XF24 Consumables

The Seahorse XF Extracellular Flux Analyzer utilizes four types of consumables:

XF24 Assay Kit

Assay KitThe XF Assay Kit consists of a disposable, 24-well, dual-analyte sensor cartridge, calibration plate, and lid. The sensor cartridge measures OCR (oxygen consumption rate) and ECAR (extracellular acidification rate).

The two fluorophores for analyte detection are embedded in a polymer spotted on the end of each of the sensor sleeves. The OCR is determined by the amount that oxygen quenches the fluorescence of a fluorescein complex. The ECAR sensor responds to changes in proton (H+) concentration as they diffuse across the fluorophore based polymetric substrate. The negative log of proton concentration is equal to the pH. The XF Analyzer's fiber optic wave guide is inserted into the sleeves of the sensor cartridge. Light is pumped through the fiber optic bundles, exciting the fluorophores and the resultant emission is detected and measured.

The sensor cartridge contains barcode information matching it to its corresponding tissue culture plate. The disposable assay kit may be used for hours or days. Sensor cartridges are not able to analyze non matching cell culture plates.

Each sensor cartridge also contains four compound storage and delivery ports per well. A unique multi-channel drug delivery system injects compounds into each cell well on command. This allows multiple measurements to be made using a single population of cells, and increases the effective throughput of the assay plate.

XF assay kits are supplied in individual containers and are packaged six per box.

XF Assay Kits may only be purchased as part of a FluxPak.

XF24 Cell Culture Microplates

Cell CultureXF24 cell culture microplates are designed to maximize the sensitivity of the sensor cartridge measurement. The well design incorporates an initial larger opening with sloping walls that terminate into a well bottom having the same diameter as a 96 well microplate.

Three 200µm high molded stops on the bottom of the plate prevent the sensor cartridge sleeves from contacting or disrupting the cell monolayer. Cells are not affected by the measurement process and can be reused.

A unique feature of the Seahorse XF24 microplate is, unlike other 24-well plates, they are designed to work with commercially available 96-well liquid handling systems.

XF24 cell culture microplates are tissue culture treated for optimum adherence and growth of mammalian cells. The 24-well plates are supplied in individual gamma irradiated containers, and are packaged ten per box. They can also be purchased as part of a FluxPak.

Three types of XF24 Cell Culture Microplates are available:

XF24 V7 Cell Culture Microplates: standard microchamber volume and cell culture surface, designed for most cell types and applications.

XF24 V28 Cell Culture Microplates: larger microchamber volume, designed for highly aerobic cells. The larger microchamber increases the dynamic range of the OCR measurement for these cell types, while the ECAR sensitivity is dampened.

XF24 V7 PET Cell Culture Microplates: molded from Polyethylene Terephthalat [PET]. PET does not absorb CO2, so PET plates equilibrate faster and may provide improved ECAR measurements.

XF24 Islet Capture Microplates

Cell CultureXF24 islet capture microplates are designed to assay the cellular metabolism of pancreatic islets with an XF24 Analyzer.

The Islet Capture Microplate uses a unique capture screen to keep islets an optimal distance from the XF sensor while permitting the diffusion of compounds and allowing for the free exchange of nutrients and gasses to keep cells healthy throughout the assay.

The XF24 Islet Capture Microplate is fully compatible with the XF24 Assay Kit sensor cartridge, making it possible to run all types of XF assays with islets.

XF24 Islet Capture Microplates are supplied with 25 capture rings n individual gamma irradiated containers and are packaged six microplates per box.

Cell CultureXF Calibrant Solution

XF Calibrant Solution is optimized for XF sensor cartridge calibration. The calibrant is pre-mixed and ready to use. A 500ml bottle will calibrate 18 sensor cartridges. The calibrant should be stored at 4°C. The calibrant solution can be purchased individually or as part of a FluxPak.

Cell CultureXF Assay Medium

XF Assay Medium is specially formulated for XF assays. Just add the required amount of glucose and adjust the pH. This ready-made, unbuffered DMEM formulation saves hours of your time. Each Seahorse certified bottle is supplied with the buffer capacity information which will improve your data quality, while lot consistency decreases variability. XF Assay medium is supplied as sterile in 1 liter bottles and packaged two bottles per box.
